Sahtoe et al. uncover how two evolutionarily related DEUBAD domains in RPN13 (ADRM1) and INO80G (NFRKB) can either activate or inhibit UCH-L5. These remarkable regulatory modes exploit flexible structural elements in UCH-L5 to modulate activity.
